Background:
Lentiviral gene therapy has been widely used in treating a wide range of genetic disorders with few approved by the FDA, such as SKYSONA®, and recently used for in vivo CAR T. Lentiviral vectors (LVVs) can package up larger foreign DNA than AAV to deliver both dividing and slow or non-dividing cells (e.g., stem cells, neurons, muscle cells). HIV Gag is one of components for lentiviral packaging and encoding structural proteins, such as capsid protein p24, which is widely used to titrate lentiviral vectors for LVV quantification.
Applications:
This kit is developed to quickly titrate your lentiviral vector samples in high throughput (~2.5h). The test is both species and isotype independent. It is for research use only.
Description:
These kits were tested to titrate a large panel of lentiviral reporters or controls, see figures for examples with catalog and calculated p24 amount. The detection linear range is 0.05-35 ng/ml; Sensitivity is 50 pg/ml.

Assay Principles (Take neutralizing antibody as example):
This kit is developed for a sandwich ELISA format to specifically capture lentiviral vector and then quantified against the p24 standard curve. The kit consists of detachable 96-well plate pre-coated with HIV-1 p24 capture molecules, p24 protein, HRP labeled secondary antibody, TMB and sample dilution/wash/stop buffer.
Brief protocol (detailed protocol is provided with the product):
- Add lentiviral samples and p24 protein standard into pro-coated plate (or strips based on sample size). Incubate at 37°C for 1 hour.
- Wash wells. Add HRP labeled secondary antibody to wells. Incubate at room temperature for 1 hour.
- Wash wells. Add TMB and stop solution. Read absorbance (OD) at 450 nm. The OD Value reflects the amount of p24 in the samples, which is then quantified against p24 standard curve to calculate titer.
Storage and shipping condition:
Shipment with room temperature. The unopened kit is stable for 6 months from the date of manufacture when stored at 2°C to 8°C. Avoid light. The shelf life is 60 days from the date of opening.
