Description:
A recombinant trimeric form of the spike glycoprotein (S protein) of MERS-Cov (GenBank: JX869059.2) was produced by human embryonic kidney HEK293 cells, followed by purification. This product contains ectodomain of S protein with S1/S2 cleavage site knocked-down, and also had a pair of mutations (V1060P and L1061P), a HRV3C cleavage site, T4 foldon trimerization domain, C-terminal 8 hexa-histidine tag and twin-strep tag. Twin-strep tag can be used for streptavidin labeling. Amino acid sequence is available upon request. This product has been run on SDS-PAGE with reduced condition and Native blue gels, with results proving trimerization and glycosylation. We also evaluated the binding of MERS-specific mAb (clone RV45, Cat. No. B73K78) and published MERS27 antibody in ELISAs, see results.
