Lenti_Pseudotyped Chikungunya Virus E with Luciferase Reporter (LR2006_OPY1)
Catalog Number:
H90Y16
Description:
This pseudotyped virus uses lentiviral vector to carry the full-length Chikungunya virus (CHIKV) structural Envelope (E) glycoprotein (Strain of LR2006_OPY1; NCBI: ALD83794.1). Chikungunya virus (CHIKV) is a mosquito-transmitted Alphavirus that causes epidemics of acute and chronic musculoskeletal disease (such as 2025 in south China). The structural Envelope (E) glycoprotein (CHIKV-E) is the major target of vaccine or neutralizing antibody development.
Quality Control:
The level of p24 on lentiviral vector determined by Quick Lentivirus Quantitation Kit (Cat. No. Q89H22) is > 1 µg/ml. Infection of cells with this pseudotyped virus carrying luciferase reporter results in high level luciferase activity. See our titration result showing that the starting 2-fold diluted pseudovirus generates signal ~ 100,000-fold higher than uninfected control (cell alone as background). Anti-Chikungunya Virus E Neutralizing Antibody (E1-specific) (clone RV78, Cat.No. Y18D21) was tested to neutralize against this pseudovirus. See picture.
Applications:
- Anti-Chikungunya virus neutralizing antibody screening at high throughput
- Anti-Chikungunya virus drug screening at high throughput
- Chikungunya virus vaccine efficacy evaluation at high throughput
- Chikungunya virus pseudovirus transduction of target cells for viral entry, receptor recognition, cellular tropism and functional studies, such as ADCC analysis
Biosafety Level:
Handle it in biosafety cabinet in BSL-2. Contacted tips & tubes should be decontaminated by 10% disinfecting bleach
Organism:
Chikungunya virus
Storage and shipping condition:
Shipping with dry ice. Require -80°C storage. Multiple freeze/Thaw cycles will reduce its sensitivity. Recommend only 1 cycle. Aliquot after the first thaw
Size:
200 μl with recommended 50-fold dilution. We recommend to use it at a dilution fold where the signal of RVP infection is 100-1,000-fold higher than uninfected control (cell alone as background). The vial is sufficient for 200 reactions (50 μl diluted virus).
Use protocol:
A complete protocol is supplied with the shipping of product.
Briefly, incubate 50 μl diluted virus with your sample in each well of 96-well plate for 60 min at 37°C. Then add 50 μl target cells, such as 293A or others expressing CHIKV receptor, followed by luciferase signal reading.
Note:
- Pseudoviruses are intended for Research Use Only
- A positive control mAb is included with pseudovirus purchase
