Description:
Mouse CD8 alpha&beta (CD8A&CD8B) Heterodimer Protein was produced by human embryonic kidney HEK cells, followed by purification. CD8 alpha (CD8A ectodomain, Lys 28 – Asp 194, Accession: NP_001074579.1) was fused with a domain called “key domain” (proprietary technology), followed with HRV3C cleavage and human Fc tag at the C-terminus. CD8 beta (CD8B ectodomain, Leu 22 – Thr 175, Accession: NP_033988.1) was fused with a domain called “lock domain” (proprietary technology), followed with HRV3C cleavage and 6 hexa-histidine tag at the C-terminus. The binding between key domain and lock domain generates heterodimer of CD8 alpha&beta. The purity has been examined by reducing SDS-PAGE, where protein with the predicted MW of 30 kDa (CD8 beta) and 47 kDa (CD8 alpha) migrates ~ 55 kDa and ~90 kDa. We evaluated its binding to anti-His tag antibody (clone RV7, Cat.No. J96C86), a CD8A-specific antibody (clone RV55, Cat.No. C94Y89), and a CD8B-specific antibody (clone RV56, Cat.No. E8V32) in ELISAs. This protein simultaneously binding to these antibodies validates the heterodimeric conformation. The protein can be cleaved using HRV3C for tag-free.
