Description:
A recombinant trimeric form of the spike glycoprotein (S protein) of SARS-Cov-2 (GenBank: MN908947) bearing a mutated S protein from D (Asp) to G (Gly) at site of 614 was produced by human embryonic kidney HEK293 cells, followed by purification. This product contains ectodomain of S protein with S1/S2 cleavage site knocked-down, and also had a pair of mutations (N1067P and L1068P), a HRV3C cleavage site, T4 foldon trimerization domain, C-terminal 8 hexa-histidine tag and twin-strep tag. Twin-strep tag can be used for streptavidin labeling. Amino acid sequence is available upon request. This product has been run on SDS-PAGE with reduced condition and Native blue gels as well as SEC, with results proving trimerization and glycosylation. We also evaluated the binding of two SARS2 Positive control mAbs (RBD-specific clone RV57, Cat.No. F5L83; NTD-specific clone RV94, Cat.No. H28V55) and CR3022 antibody in ELISAs, see results.
