Description:
This antibody was derived from a human survivor of the 1995 Kikwit outbreak of Ebola, and binds to the Ebola virus surface glycoprotein in its trimeric, pre-fusion conformation (GP1+GP2). KZ52 recognizes a protein epitope at the chalice base where it clamps several regions of the pre-fusion GP2 to the amino terminus of GP1. This antibody neutralizes Zaire ebolavirus in vitro by preventing rearrangement of the GP2 HR1A/HR1B segments and blocking host membrane insertion of the internal fusion loop. Alternatively, IgG KZ52 may sterically hinder access to the RBS or to a separate binding site of another cellular factor, especially if multiple attachment events are required for entry (Lee et al. 2009).
