Background:
Fluorescence-activated cell sorting is a type of flow cytometry for sorting a group of cells with specific light scattering and fluorescent characteristics. The cell sample is stained with fluorescence antibodies for cell surface markers and entrained into a nozzle to form droplets, followed by laser-based fluorescence analysis. The fluorescent character of interest of each cell is electrical charged and sorted by the opposite charge on the deflection plates. The sorted cells are phenotype defined with great homology. The sorted cell can be collected in single or bulk format.
Applications of sorted cells:
- Antibody development: Human memory B cells are widely sorted from PBMCs and defined as CD3−CD8−CD14−CD20+IgG+CD27+IgM−. These memory B cells can be further defined by one or more probes for BCR-specific, e.g. HIV-1 Env where HIV neutralizing antibody VRC01 was isolated (Ref: Wu etc, Science. 2010; 329: 856–861), or SARS-CoV-2 Spike where COVID-19 therapeutic antibody REGEN-COV® (casirivimab and imdevimab) were isolated (Ref: Hansen etc, Science. 2020; 369: 1010–1014).
- Vaccine development: The sorted cells with vaccine antigen specificity are useful to evaluate immune response.
- Tumor cell profiling for oncology: the sorted cells with cancer cell specificity are crucial for understanding tumor progression, metastasis and response to therapies. It facilitates the biomarkers’ identification for diagnosis and treatment development.
- CAR-T cell therapy: the isolation of primary T cells from donors is critical step for the success of CAR-T therapy.
- Stem cell research: the sorted stem and progenitor cells from the mixed cell population facilitate the study of stem cell differentiation pathways and potential for medicine development.
- Signal pathway and apoptosis: the sorted immune cell or T cells for signal transduction or cellular apoptosis detection contribute to study on disease mechanism and immunotherapy development.
Our service includes:
- Incubate cell sample with fluorescent conjugated primary antibodies following the provided protocol.
- Incubate bead with fluorescent conjugated primary antibodies for compensation.
- Run above mixture on BD FACS Aria Cell Sorter (5 laser system) to gate and sort as requested.
Service return:
- The collected sorting products (single or bulk cell with the defined specificity) in vessel as requested.
- Real-time gating/sorting report in pdf file.
- FACS fcs files.
Service turnout:
The Services are usually completed within 1 week after the following received.

The listed price is per sample you request to sort. Please choose based on your request of cell collected in single or bulk format.

- Collecting vessel with buffer or culture media
- A protocol
Note:
- We only accept samples handled under BSL-1 or -2 containment, not BSL-3 or -4.
- We can provide fluorescent conjugated antibodies/beads/collecting vessel if you don’t have. But it will raise the cost.
- We would like to provide suggestion for protocol upon request.
