Description:

This Seamless Assembly Master Mix (2X) uses Gibson cloning principle to allow efficiently assembly of multiple DNA fragments, regardless of fragment length or end compatibility. The insert and the linearized vector need the overlapped sequences of 15 bp – 20 bp on both 5′ – and 3′ -end, respectively. A positive control with the mixture of insert and linearized vector is included.

Features:

• Assembly is a one-step mixture with the incubation up to 1 hour at 50 °C
• No restriction enzyme digestion, gel extraction or overnight ligation
• PCR clean-up step is optional, even clean-up will increase cloning efficiency
• Adjacent DNA fragment needs shared overlaps
• High transformation efficiencies for inserts up to 20 kb
• Easily manipulation of up to 4 DNA fragments into linearized vector
